They are also critical for the virulence of many pathogenic bacteria. Type-III secretion systems (T3SS) enable the assembly of two homologous nanomachines: the bacterial flagellum and the injectisome. This process, relies on a protein complex called the export apparatus to coordinate the highly ordered assembly of tens of different substrates, which are recognized and injected at high speed across the bacterial cell envelope.
This high speed and versatile secretion process can be harnessed for biotechnological purposes. By engineering the bacteria, it is possible to drive the specific secretion of exogenous recombinant proteins. In the laboratory, we have identified and modified natural substrates to be excellent drivers of exogenous proteins secretion with expected industrial and medical applications.
The proposed research project will have both fundamental and applied perspectives. We will aim at deciphering some of the basic mechanisms of type-III secretion, in particular related to the recognition and efficiency of secretion of natural and artificial substrates.
Furthermore, we will engineer the genetic background of the bacteria and the secretion nanomachines to develop candidate biotechnological applications.
