Offer Description

In the context of the ATHENA EU H2020 project https://athenaedctestmethods.net/ ,
the short and long term effects of perinatal exposure to thyroid hormone axis disuptors, alone or in mixture, will be studied on two relevant models, mouse and amphibian.
The candidate will study effects from molecular/cellular levels in brain tissue to behavioral endpoints.
He/she will be also study consequences of maternal thyroid status on the impact of endocrine disrupting compounds exposure
Creation of crispants amphibians, mimicking hyperthyroxinemia or hypothyroxinemia are planed, especially KO for thyroid hormone (TH) transporter.

On two relevant and complementary models, effects of a developmental exposure on brain development are foreseen. Indeed the candidate will analyse consequences of a developemental exposure with a common mixture of endocrine disruptors is planned. Collection of tissues, cell proliferation, differentiation analysis and behavioural analysis are foreseen. Transcriptomic analysis (RNA extraction qPCR, RNAseq) are also planned.
Regarding histology , immunohistochemistry and Confocal imaging are possible
Regarding behavioral analysis, Software and dedicated equipment are accessible
CrisprCas9 genomic invalidation support is accessible.

PhyMA is a Joint Research Unit of the Muséum National d’Histoire Naturelle associated with the CNRS (UMR 7221). It is interested in the physiology of organisms at all scales, from their cells to their environment.
It is composed of 3 research teams, and includes 50 people (permanent and non permanent). This unit is located at the Jardin des Plantes in Paris 5th. The person recruited will be affiliated to the RODEO (Response to envrionmental challenges) and BBC teams (Brain-body crossalks) under the direction of Dr Sylvie Remaud and Pr Jean-Baptiste Fini.

Additional Information
Eligibility criteria

– Solid knowledge in endocrinology (thyroid physiology would be a plus) and in molecular biology
– Knowledge of endocrine disruption, neurodevelopment in vertebrates
– Skills in data analysis
– Initial experience including immunohistochemistry techniques would be a plus.
– The candidate is willing to be well organised, autonomous and highly motivated.
– Bioinformatic skills would be a plus.
