Un projet financé en 2017 par l’ED MTCI a permis d’identifier le premier « hit » synthétique d’un alcaloïde non-développable comme antipaludique, la tazopsine. Le projet de thèse proposé vise une continuité de développement thérapeutique de ce « hit » simplifié, accessible et d’activité antiparasitaire déjà vingt fois supérieure au produit naturel. L’approche prévue est une diversification par chimie « clic » combinatoire en plaque 96 puits, suivie d’une évaluation biologique et optimisation physicochimique des « early leads ».

A project funded in 2017 by ED MTCI identified the first synthetic «hit» of an alkaloid non-developable as an antimalarial, tazopsin. The proposed thesis aims at a continuity of therapeutic development of this simplified and accessible «hit», already showing an activity twenty times higher than the natural product. The planned approach is a combinatorial 96-well «click» chemistry diversification, followed by a biological evaluation and physicochemical optimization of «early leads».

Specific Requirements

– Posséder un M2 “Chimie Organique” (synthèse totale, méthodologie de synthèse ou chimie médicinale +++).

– Avoir une mention “Bien” en M2 (impact très important sur la sélection des candidatures par l'ED pour la session orale)

– Expérience / intérêt en biologie: le/la candidat(e) devra être capable de réaliser une thèse à l'interface sur 2 pôles parisiens (chimie à la Faculté de Pharmacie, tests biologiques à l'UPMC), proches géographiquement et où il/elle sera formé(e) par des équipes expertes et partenaires depuis 10 ans.

– Have an M2 (or equivalent) in “Organic Chemistry” (total synthesis, synthetic methodology or medicinal chemistry +++).

– Have a “Good” mention in M2 (very important impact on the selection of applications by the ED for the oral session)

– Experience/ interest in biology: the candidate must be able to complete a thesis at the interface on 2 Parisian poles (chemistry at the Faculty of Pharmacy, biological tests at the UPMC), geographically close and where he/she will be trained by expert teams and partners for 10 years.
