Offer DescriptionThe postdoctoral project is part of the ChromSwitch project funded by the ANR, which aims to study a function that releases Polycomb PRC2-mediated chromatin repression of developmental genes in plants.
The main objectives of the postdoctoral project are (1) to characterize further a PRC2-associated factor, carrying this switch function, including its molecular features; and (2) to resolve its interaction and opposing mechanism with PRC2.
For this, the researcher’s mission will be to develop molecular and plant genetic approaches and tool for epigenomics/epigenetics and interactomics analyses.The post-doctoral researcher will develop a project under direct supervision of C. Carles (team leader) and G. Vachon (researcher in the team). The post-doc will benefit from the team’s network of collaborations in Grenoble, within the ChromSwitch ANR consortium. In particular, the researcher will work in close interaction with the permanent engineer in charge of molecular and plant work (M. Le Masson), as well as our partner at the Institute of Structural Biology (IBS).Activities will include:

  • Genome-wide profiling of epigenetic marks and proteins: ChIP-seq, Bisulfite-seq
  • in vitro Protein-DNA interaction analyses: EMSA, fluorescence anisotropy, Surface Plasmon Resonance
  • in vivo functional assays: Luciferase assays (protoplasts), mutant complementation analyses (Arabidopsis thaliana)
  • Interactomics: IP-MS, ChIP-MS, Split-BioID-MS; Y2H, BiFC or Split-Luciferase
  • in cyto immuno-localization

In addition, the post-doc will present data in English at team meetings and laboratory seminars, as well as participate to the analysis of scientific articles related to the researcher’s research, and discuss them with team members (e.g. in journal clubs).The researcher will be part of the ChromDev team ( ) of the Cellular and Plant Physiology Physiology Laboratory (UMR 5168 – CNRS -CEA – Univ. Grenoble Alpes – INRAE) in Grenoble. The team’s projects aim to better understand the Chromatin Dynamics that regulate Developmental Transitions in plants, using a multi-disciplinary strategy, encompassing in vivo phenotypic analyses, omics studies (epigenomics, transcriptomics, interactomics) and in vitro characterisation of molecular interactions and 3D structures of protein complexes.This position will take place on the Polygone campus in Grenoble, a rich environment for biology research, close to several laboratories such as IBS, IRIG, ESRF and EMBL. The LPCV institute employs around a hundred people and boasts state-of-the-art equipment for molecular biology, biochemistry, structural biology and imaging. It has plant culture facilities where all steps of molecular genetics on Arabidopsis can be carried out. The candidate should :
– master the basics (theory and practice) of genetics/epigenetics and molecular biology.
– be familiar with methods for analyzing and measuring gene expression and epigenetic marks, and their use in bioinformatics.
be familiar with methods for bioinformatics analyses and measurements of gene expression and epigenetic marks.
– be autonomous in designing experimental plans, interpreting and presenting results at team meetings and seminars
– develop tools and methods and use them in a reliable and reproducible manner.
– be able to contribute to the preparation of scientific publications.
– be able to work independently in an interdisciplinary and international environment.
– be willing to interact scientifically in a dynamic research group.
– respect laboratory rules and procedures.
