The research project focuses on developing innovative strategies for modulating polyamine metabolism by inducing molecular proximity between proteins that are key to this process. The project aims to develop new molecules that induce proximity in the polyamine metabolism pathway by combining methodologies such as crystallographic fragment screening, imaging of protein complexes, and studies of cellular polyamine metabolism. The goal is to create a bifunctional molecule that stabilizes the complex of ornithine decarboxylase (ODC) and antizyme (Az), which are crucial for polyamine metabolism. Additionally, the project aims to develop the first-in-class modulator of the hypusination process, a unique modification of the eIF5A factor essential for protein translation. Structure-based studies of protein-molecule complexes and their chemical optimization are key to the project’s success. The experiments will be supported by data analysis from various research techniques, including cryo-electron microscopy. The studies will be validated at the enzymatic and cellular levels to assess the efficacy of the developed molecules. The project will contribute to the development of novel strategies for modulating polyamine metabolism and the creation of new research tools with therapeutic potential.Scope of duties:According to the Work Regulations of the Jagiellonian University Annex 1 to the Work Regulations of the Jagiellonian University – Model scopes of responsibilities and duties of academic teachers.The hired person will be responsible for independently coordinating tasks, making rational and logical decisions to solve problems, and introducing new techniques and tools. The best-selected candidate will be required to learn and optimize new methods in the fields of structural biology, biochemistry, biophysics, and medicinal chemistry.   • holding at least a doctoral degree;
  • having relevant scientific achievements;
  • taking active part in scientific life.

Specific RequirementsAdditional requirements and expectations:Ideal candidates:

  • have relevant scientific background in cell biology, molecular biology, biochemistry and/or related fields,
  • have at least one original scientific publication, in which they are the lead author,
  • take an active part in scientific life manifested in particular in presentations at conferences and symposia
  • meet the FNP requirements for persons employed in research projects as a Young Doctor (up to 7 years after obtaining a doctoral degree ( )

In addition, the candidate is expected to have prior knowledge and experience in at least one of the following of the listed methods:

  • Experience in research in the fields of structural biology (X-ray crystallography or cryo-EM), biochemistry, biophysics, protein-protein interaction studies, drug design, screening techniques, peptide design, DNA/RNA aptamer design, molecular biology, biophysics, and related topics.

The ideal candidate will possess:

  • Knowledge of recombinant and native protein purification techniques.
  • Knowledge of molecular biology methods such as cloning, expression construct design, Western blot, RT-PCR.
  • Knowledge of prokaryotic and eukaryotic protein expression systems.
  • Experience in studying protein activity using various enzymatic assays.
  • Experience in macromolecule structure analysis.
  • Experience in the discovery and/or design of bifunctional compounds, such as those from the class of molecular glues.
  • Experience in synchrotron measurements or cryo-EM.
  • Experience with methods such as SPR, HPLC, MST.
  • Proficiency in English at a communicative level, enabling daily work in an international scientific environment, presenting results, and preparing scientific articles.
  • Familiarity with basic MS Office programs and optionally with the Linux operating system environment and software such as CCP4, Phenix, Relion, CryoSpark, PyMol, GraphPad/Origin.
